Setting Data Scaling Preferences

Select the Data Scaling button in the Preferences tool to open the Data Scaling preferences. Roll over the red dots for more information.

Please note: this preference panel is meant to set a starting point to scale your digital FCS3 data. Fine-tuning adjustments (if necessary) should be done using the T button in the Graph window.

For your convenience, below you can find examples of what numbers to use in this panel for high resolution digital files such as the 5-decade BDFACSDiVa and Accuri C6 files.

 

For DiVa data with and without Transforms, use these settings:

If you choose to display data in biexponentially transformed plot (recommended by this Herzenberg paper) - the -100 value of Width Basis will give a nice compression around 0, giving the negative population a rounded appearance:

However, if you need to view this data file without transforms, the high starting linear channel number will trim out the picked-fenced first decade and make the plot appear more like old FCS2 style data:

 

For Accuri C6 data, you may want to increase starting channel number to 1000, and use the same value for width basis if you will be transforming the plots.
