There are two types of Compensation Wizards (CompWiz) in FlowJo 7.5: a Stepwise and a Drag-and-Drop.
The Stepwise CompWiz
Prior to creating your matrix, we recommend always creating a group that contains only your compensation controls. For more on Groups, click here.
Parameter Label Confirmation:
Once your group has been created, select Compensation Wizard... from the File menu of the Compensation Editor. The StepWise CompWiz will open on the Parameter Label Confirmation screen and FlowJo should identify your single stain controls. If your comp control group is selected in the Workspace then click on the Use Current Group button, otherwise confirm your parameter labels by selecting the control samples individually from the drop down menus.
When you have confirmed your parameter labels, click Next.
Gate Confirmation:
On the gate confirmation screen you'll see the plots and gates for autofluoresence and positive and negative populations that FlowJo has generated. Positive gates are on the left side of the window, negative are on the right. To adjust a gate, double click on its plot. The plot will open in a separate graph window where you can make the needed adjustments. If FlowJo was unable to find a suitible gate, you will be notified that of the missing population. Double click on the notification to get a graph window where you can create the gate.
Once you have confirmed your gates are properly placed, click Next.
View Matrix:
On the View Matrix screen you will see the full matrix that FlowJo has calculated based on the two prior steps. Once you have confirmed the values of the matrix, click Finish. You will be returned to the Compensation Editor, where your new matrix will show. You can then assign samples to the matrix or edit and save the matrix.
Go to the Overview of Compensation.
